Abstract: Immunological approaches are based on the specific reaction of antibodies with an antigen. They are suitable for the identification of proteinaceous binders and offer several advantages over the traditional methods used for analyzing binding media. These techniques are able to distinguish between different proteins (i.g. collagen, albumin, casein) but also to determine unambigously the species (i.g. bovine collagen, rabbit collage, sturgeon kollagen). In this context two methods should be mentioned: The Enzyme-linked Immunosorbent Assay (ELISA) is highly sensitive (detection limits below one nanogram) and relativly simple, whereas the Immunofluorescence method (IFM) offers the possibility of locating the target antigen (protein) in embedded cross sections. In this study it was possible to detect and identify casein, ovalbumin and sturgeon collagen in fresh samples whereas the detection of binding media in aged samples seems more difficult. Cross reactions and possible inhibitors like metal based pigments or wood preservatives were tested on the influence on the immunological reaction. Both techniques, the ELISA and IFM show potential of the use for the analysis of binding media. With further research and standardisation they could become a usefull tool in conservation science.
